杨树是世界主要的造林和经济树种,在中国和世界各地均有广泛的栽培?格林一号黑杨(G1)是一种欧美杂交杨,具有生长速度快?材质好?适应性强等优点,为目前推广的主要品系之一?本研究选择G1黑杨作为转基因的实验材料,研究了激素浓度等因素对其叶片再生?壮苗?生根等的影响,建立了杨树高频再生体系?几丁质和葡聚糖是许多病原真菌细胞壁结构的重要组成部分?几丁质酶可以将几丁质水解为N-乙酰氨基葡萄糖;β-1,3-葡聚糖酶可以将葡聚糖降解为葡萄糖?大量研究表明,以上两种酶在植物体内体外,对病原真菌的生长均有明显的抑制作用?本研究将这两种酶的编码基因分别构建植物表达载体,以G1黑杨幼叶为外植体,利用改良的农杆菌介导法进行转化,获得转基因植株?具体实验结果如下:1。

杨树是世界上中纬度地区广泛栽培的重要用材树种,具有广泛的工业用途,世界上许多国家把发展杨树作为解决 杨树是世界主要的造林和经济树种,格林一号黑杨(G1)是从欧洲黑杨与美洲黑杨的杂交后代中选育出的一个无性系,具有抗逆能力较强?材质好和速生等优点,在我国北方广大地区既可作为绿化树种,又是制作高级纸的主要纸浆原料之一,在工业用材中发挥重要作用? 本研究以 G1 为实验材料,以其叶片为外植体进行再生培养,系统地研究了激素浓度?叶片生理状态?光照条件等诸多因素对叶片再生的影响,建立了黑杨叶片培养诱导不定芽的高频再生体系?在此基础上,分别用携带 BG2 基因和反义磷脂酶 Dγ基因(PLDγ)的根癌农杆菌介导转化叶片,通过对转化方法和条件的探索,建立了黑杨 G1 的遗传转化体系? 研究结果表明,不同激素浓度配比和 pH 值对芽再生有重要影响,最终以在 MS 基本培养基中附加 0。
